ATHLETICS. The Russian Anti Doping Laboratory: the structural properties of the molecule of 6-alfa-Methyl-Androst-4-en-3,17-dione do not allow to classify it as a prohibited substance and doping

The structure of 6-alfa-Methyl-Androst-4-en-3,17-dione, for which the Russian athletes in hammer throwing Tatyana Lysenko and Ekaterina Khoroshih have been suspended from competition, can not be considered as a doping without further elucidation and appropriate consideration. It follows from the explanatory letter from the Moscow Anti Doping Center. The letter was requested by All-Russian Athletics Federation (ARAF). The Agency of sport information "All Sport" publishes it without changes.

Dear Oleg Zaharovich,
In reply to your request made on August, 3, 2007 about the metabolite of 6-alfa-methylandrostendione found in the organism of the athlete Tatyana Lysenko and the problem of classifying it to the List of the prohibited substances we can inform you on the following.

This compound is not included in the Prohibited List of the World Anti-Doping Agency (WADA) for the year 2007 and it is not included in the draft of the List 2008. None of the athletes, as far as we know, has been disqualified for this substance. But the WADA list in the part of the anabolizing (chapter S1) and the anti-estrogen agents (chapter S4) is not exhaustive, that means that the other substances with the similar chemical structure and biological effect may be attributed to them.

According to its structure the substance 6-alfa-Methyl-Androst-4-en-3,17-dione can not be unambiguously classified as the anabolizing agent, since the anabolic steroids usually have methyl substitution at the positions 1, 2, 7, or 17, and there is no one anabolizing agent in the current List with methyl group at the position 6. Then, the possible classification of this substance to the group of the anti-estrogen agents also questionable. Structurally similar analog with anti-estrogenic properties is exemestane (6-Methylene-Androsta-1,4-dien-3,17-dione), which has methylene group at the position 6, but it differs in having two double bonds and planar structure of 6th position. It is important to mention that minor changes in the structure of steroid molecule can significantly change its biological effect on the organism.

Therefore, the classifying of 6-alfa-Methyl-Androst-4-en-3,17-dione as anabolizing or anti-estrogen agent should be based on the scientific data on its biological effects, because the structure of the molecule does not allow unambiguous classifying this compound to the prohibited doping substances. I have sent the inquiry to the Scientific Center of the Expertise of the Medical substances with the request to make the expert conclusion about the medical effects of this substance and provide scientific data necessary to classify the substance in question as a medicine with the anti-estrogen activity.

The famous Russian runner, 800 m Olympic champion Yury Borzakovsky was found not guilty in the car accident that happened at night September, 30 – October, 1 in Zhukovsky, Moscow region. The car accident resulted in a man’s death but the criminal case was not even started.
